Check out the Western Flyer or the Tri-Star from Tom Bihn. Both of these can accept a laptop insert and they can be carried or worn as backpacks. I have a number of his bags--they are all thoughtfully designed down to the last detail, incredibly durable and made in the U.S.A.

While he doesn't have any bags that can really double as both a work bag and an overnighter, Jack Spade has been making great looking and durable bags for years. I can't recommend the Spire Torq enough. It's extremely durable, capacious, and specifically built for carrying a laptop around. Definitely roomy enough to pack for a night, and comfortable too. Their bigger Meta is massive, but the main compartment can handle a pair of shoes and a folded garment bag. with no issues.
