It depends what kind of liquor you like (vodka, rum, tequila, whiskey, bourbon, scotch, or gin), I can give you my personal favorite for each:

I'm going to say a Bourbon and Ginger or a classic Manhattan with Knob Creek Bourbon (9 years aged, pretty smooth and readily accessible)
I don't know if these are manly or not, but I do enjoy them. If I'm in the rye/bourbon type mood; Sazerac, Manhattan, Rob Roy or an Old Fashioned. A Gin Gimlet if I'm up for some gin. A quality single malt or blended scotch is always appropriate. And too, you can enjoy your Rob Roy with a bourbon or a blended scotch depending on your mood.

Mixed drinks can be delicious, and certainly worthwhile, but I don't know if "manly" is an appropriate adjective. If one must mix, consider mixing scotch, or bourbon, or Jack Daniels (my personal favorite) with ice.

Mixed drinks? Real men drink their liquor straight! I prefer a solid whisk(e)y, usually bourbon or scotch, with a splash of water to open up the flavor. If you must go with a mixed drink, though, Greg Briere's on the right track. I'd recommend a Manhattan or, if you find yourself in more of a dive, a simple bourbon and ginger.
And if you're lucky enough to live near a place like Drink in Boston or the Gibson in DC, which elevate mixology to an artform, you might spend some time getting to know the bartenders there. They'll show you more than a few amazing manly mixed drinks.
While straight liquor is plenty manly, there's something about being particular about "shaken, not stirred" that denotes true manliness.
I'd recommend the Sazarac. As the oldest, and first, cocktail invented in America, there's really something badass about drinking 1830's-era Rye Whiskey and Bitters.
Colorado Bulldog, Alabama Slammer, Grateful Dead, Red Death, Godfather, Madras (pronounced muh-DRAHS so you don't sound like a cretin), Manhattan, Old-Fashioned, Sidecar, Rob Roy, Black Russian. All delicious, manly sounding drinks (which is what you're really going for, right?)
If you're not in the mood for a multi-ingredient cocktail, there's nothing wrong with playing it safe and ordering something simple like a vodka tonic, bourbon ginger, rum and coke, Red Bull and vodka, etc.

I'm definitely a whiskey drinker (irish for the most part). Rarely ever mix it with anything.
My mixed drinks of choice are a dirty goose up, goose and soda with a twist of lime, and the occasional margarita on the rocks never with crushed ice.
Manly mixed drinks are simple things, what I would call "& drinks", as in booze & mixer.
For example: whiskey & coke, gin & tonic, vodka & red bull, etc.
Keeping it simple helps keep it manly. Of course, you can always stray accidentally and unknowingly. If you order a vodka and cranberry, it follows the rule, is every bit as strong as any other listed, and tastes good. But it's not really manly.
Maybe there should be a corollary, if it's fruity, it's not manly. Screwdriver falls into that category. I don't think it's a manly drink at all, even though it started as one.
A gimlet, as mentioned, I would consider manly, even though it has "fruit". But it's lime, so it's not sweet, it's tart.
If you need sweet, I think a 7&7 might be ok. It doesn't look fruity because you have a clear mixer.
Is that another corollary? Avoid bright colors in your mix. No orange, definitely no pink, no red, no bright greens. Keep it clear or brownish and I'll think you'll still be manly.

I'd imagine it is a mixed drink that you may have seen someone you consider 'manly' (father, uncle, grandfather, etc.) drink when you were young.
For me, that would be rum & coke, jack & coke, or gin gimlet. All of those were tried out in my younger days, when the drinking age was still 18.
Personally I don't care for any of them, I prefer scotch. Because I am not rich, it's Johnnie Walker Black on the rocks. I just let the ice melt for 3-5 minutes or as long as I can hold out. I even drink it in the summer. For shame, I know.
The single malts that I have liked are expensive, and often not found at your average bar.
I really don't consider Bloody Mary's a mixed drink...I think of (and enjoy) them more as breakfast.

If it's got more than two ingredients, it's not manly. And girls drink out of straws; boys don't.

I've been a sailor for 20 years and I've never had a Dark & Stormy.
Also a martini is 2 ingredients, to wit:

shake w/ice
strain into glass
add olive

Can't beat the way Belvedere and fresh grapefruit juice mix.
Any drink can be manly depending how you order it. Be confident. Get what you like. People frequently switch to Greyhounds after I order one (it's all about the juicer).
Normally I drink Pabst Blue Ribbon, on the rare occasion I get a mixed drink it's rum and coke or a whiskey sour.
My personal preference is a good top shelf-ish bourbon like Knob Creek. Anything from the Beam distillery is good, even plain ole Jim Beam. Jack Daniels is, in my humble opinion, nasty rotgut toilet water that helps sort out people who are trying to order something that sounds manly, from someone who appreciates the flavor of bourbon on ice. If someplace doesn't have bourbon, beer is better than whiskey. PBR tastes like the can, but it is a cool looking can.

A margarita, whiskey sour, anything mixed with Coke, etc. should not be considered if we are asking the question, “What is a man’s drink?” While the Dark & Stormy, Margarita, etc. are good cocktails, when made properly, they are not what a discerning gentlemen would order on a Friday night, at say a steak house. Never order a Coke and anything unless you’re in jeans shorts or using your fake ID.
Whiskey, specifically bourbon or rye, is arguably the quintessential “man’s drink.” Pulp fiction detectives, astronauts, and rock stars have been associated with good, strong whiskey. As some people have mentioned, bourbon or good rye on the rocks will do the trick. If you don’t like it straight, first check you’re drinking something decent. Maker’s Mark is distilled with more wheat than other bourbons and is therefore slightly smoother and therefore a good starting point. If it’s still too strong for your taste, consider ordering it neat alongside a cold bottle of beer. If you’ve got the persona, order a “cold king with a whiskey back” – bar slang for a bottle of Bud and a shot. Finally, if the bar looks up to the task, order a Manhattan made with with rye (Wild Turkey makes a nice rye and Sazerac is not bad), if possible, and tell them to hold the cherry.

Trick question. 'Manly Mixed Drink' is an oxymoron. That said, I had an uncle (from Boston, and Harvard) who insisted on getting his Manhattan in an old fashioned glass, two maraschino cherries and no ice. The mix was 3:1 Canadian Club Rye and Sweet Vermouth and a splash of the maraschino cherry juice.
So, to honor my Late Uncle Bob, that's become my drink. I'm not as picky about the glass, but I insist on no ice. And the cherries.

IMHO nothing with Red Bull or Vodka in it is remotely manly. Unless you are a man living in a sorority house - then it's understandable, I suppose. Real men's mixed drinks are classical cocktails that enhance and celebrate the flavor of the real spirit base of the drink. Cognac, Whiskey, Gin, Rum, or Tequila are proper base spirits. Proper men's cocktails don't disguise the spirit, but stick to the classic format for a proper cocktail: one bitter, two sweet, four strong, eight weak. bitters, a sweetner, a stiff snort, and ice. Martinis (gin, not vodka), Old Fashioneds, Sazeracs, Sidecars, Daiquiris, Margaritas, etc., qualify. If Papa Hemmingway would have drunk it without embarrassment, it's a man's drink.
Manly drinks are defined mostly by what they are not. They are not trendy. They are not merely marketing for the latest bottled tipple. They do not use pre-mixed concoctions (sour mix makes a real man weep). They do not feature more juices or sodas or liqueurs than base spirits. They are not festooned with fruit.
Finally, beer and wine. Both manly, so long as you drink what you like, not just whatever is available. Champagne is manly, without a doubt, whether taken by itself or in a cocktail. The one exception is any wine or beer with a fruit flavoring in it. Real men only drink fruity beer or wine in the basement when no one is looking.
Above all, manly drinks are drunk for pleasure, not just to get a buzz on. Real men don't binge, they control their intake so as not to embarrass themselves and others, nor ever drive while impaired. Real men don't kill innocent people by accident.
After conferring with some very intelligent and outspoken women at my office, the only acceptable mixed drinks a man may order at a bar without looking feminine are as follows: rum and coke, jack and coke, vodka and tonic, gin and coke, 7 n' 7, and an irish car bomb (on St. Patrick's Day). In other words, don't get too crazy with your choice, because it can change initial perception, especially among women you may meet.

Don't mock other peoples opinions, because that is what weak men do.
It's not about what everyone else thinks is manly... it is all about what makes you feel like a man when you order it, when someone asks what you are drinking, and when you drink it. (If you are still in college, it is also important to remember to feel like a man when you are barfing it in your dorm bed) Seriously, whatever you order... you own that drink and commit to it. Don't order a crown and ginger then turn around and order a Jack on the rocks... everyone will realize you are trying too hard! Stick with the same drink all night.
My suggestion is to purchase several different types of dark liquors for the house and learn what best suits your taste. (I drink Makers Mark, Crown Royal, Jack Daniels, Jim Beam or Vodka on the rocks) Try mixing with coke, ginger ale, water, and as a last resort sprite.
Before ordering anything on the rocks... learn how to drink on the rocks.
Keep your confidence in check... 6-8 drinks into the night is not a good time to begin experimenting. Do that at home.

The only reason people feel that whiskey sour is manly is b/c whiskey is in it's name. Don't order a whiskey sour.

If you order a shot for the table order something with a dark liquor in it. Do not order a shot that has a name that refers to anything sexual... especially if you are only drinking with the boys or sending a shot to a table of girls that you do not know. Jager Bombs are always a crowd pleaser.

Stick with bourbon (no coke!) or scotch (i prefer single malt) neat. Until you can handle either of those straight, stick with non-light beer. Light beer was originally brewed for female consumption to keep revenues up while men were overseas fighting wars. No lie. Light beer is for ladies. Aqcuire a taste for the IPA's, ales, and stouts. Start out with lagers and wheat beers if you're not quite there.

A man's drink - usually straight. If you're doing Scotch, skip the rocks (it waters it down, unless you can't handle scotch, then work your way there). Vodka is a plus, and the higher quality tends to give a nice, smooth finish. Martinis have the "007" appeal, but it depends on the crowd.
It boils down to presentation - who're you tring to impress? The guys at the bar? Grab whatever beer is most in demand. If they have a good selection, go for one that you like (microbrews, seasonals, whatever).
Jesse's right about drinking straight whiskey, but ignore the "lose the cherry" with a Manhattan - that's just trying to hard. Don't try to put up a front, just find the drink that gives you your perceived edge without trying to be pretentious.

As I get older my choices are more thought out, I want to enjoy my drink.
I no longer drink solely to get drunk. Although the drunk part of drinking I do enjoy.
Good bourbon. Neat. shot of bourbon in a Old Fashioned glass (AKA lowball glass or rocks glass) no ice room temp.
Sip it. enjoy it. taste it.
Beer is always good. I tend to enjoy Guinness and or a hoppier brew.
Hoegaarden is an all time favorite. Its a little yeasty but smooth.
As far as the drink of choice being "Manly" that also depends on who you ask. What is manly here may not be there.
So use you head and your taste buds and sort it out.
